Smith, D. W. (University of Wisconsin, Madison), G. B. Fregnan, L. DeLaquerriere-Richardson, and E. Valdivia. Induction of acquired resistance in guinea pigs with defatted Mycobacterium tuberculosis vaccines. J. Bacteriol. 88:87-92. 1964.-Residues of mycobacteria exhaustively extracted with neutral organic solvents have been shown to produce a level of resistance against tuberculosis in guinea pigs comparable to that produced by BCG. Disruption of defatted tubercle bacilli in adjuvant eliminated the acid-fast staining property of the residue, but immunizing and sensitizing properties were retained in the fraction which sedimented at 44,000 x g. To demonstrate these activities with 50 mug of vaccine, it was necessary to use Bayol-Arlacel adjuvant. A similar quantity of vaccine given in saline was inactive. A defatted vaccine prepared from a virulent strain of Mycobacterium tuberculosis recently passed through a guinea pig was significantly more immunogenic than a similar vaccine prepared from H37Ra.

IMPORTANCE OF CATALASE ENZYME IN VIRULENCE OF ISONIAZID RESISTANT ST RAINS OF MYCOBACTERIUM TUBERCULOSIS IN GUINEAPIGS
In this study, twenty-five strains of Mycobacterium tuberculosis resistant to isoniazid (INH) were isolated from patients with tuberculosis (TB). Nine strains (36%) were found to be virulent in guinea-pigs [root index virulence (RIV» 1 ]. The remaining sixteen strains (64%) were non-virulent (RIV <1). Of the nine strains resistant to INH as well as virulent to guinea-pigs, eight of them wer...
متن کاملPURIFICATION AND CHARACTERIZATION OF PROTEIN ANTIGENS ISOLATED FROM MYCOBACTERIUM TUBERCULOSIS (H37Rv STRA IN) AND THEIR EFFECTS ON CELL-MEDIATED IMMUNE RESPONSES IN GUINEA PIGS
Mycobacterium tuberculosis (H37Rv strain) was used in this study. The bacterial cells were disintegrated by sonication. The separation and characterization of the soluble molecules were attempted by various techniques including gel filtration, ion exchange chromatographies and polyacrylamide gel electrophoresis, using SDS and 2ME. Eight protein molecules with molecular weights ranging from...
متن کاملVirulence in isoniazid-resistant clinical isolates of Mycobacterium tuberculosis from south India
Isoniazid, is the only antituberculous drug for which the relation between lack of virulence and acquisition of resistance was associated. INH-resistant mutants were shown to contain defective katG gene. Classical studies showed that INH-resistant south Indian isolates have lower virulence in guinea pigs and higher susceptibility to H2O2. It is of interest to assess the virulence in south India...
متن کاملLack of protection in mice and necrotizing bronchointerstitial pneumonia with bronchiolitis in guinea pigs immunized with vaccines directed against the hsp60 molecule of Mycobacterium tuberculosis.
In this study, the hsp60 and hsp70 heat shock protein antigens of Mycobacterium tuberculosis were tested as potential vaccine candidates, using purified recombinant protein antigens or antigens encoded in the form of a DNA plasmid vaccine. Guinea pigs vaccinated with a mixture of the two proteins showed no evidence of resistance to low-dose aerosol challenge infection and quickly developed seve...
متن کاملStudies of Tubercle Bacillus-histiocyte Relationships
Immunization of mice or guinea pigs with BCG rendered all or most of the histiocytes of these animals resistant to necrotization by virulent H37Rv; this cellular resistance was mediated by immune serum. Immune mouse histiocytes (from BCG-immunized animals) were able to induce cellular resistance in normal homologous and heterologous (rabbit) animal species; mouse histiocytic ribosomes were also...
